Génétique moléculaire des lactate-déshydrogénases des bactéries lactiques

Abstract - Molecular genetics of lactate-dehydrogenases from lactic acid bacteria
We have . cloned 2 D-lactate-dehydrogenase genes (D-LDH) and one o-hydroxy-isocaproate-dehydrogenase gene (D-HicDH) from lactic acid bacteria (1...actobacillus delbrueckii subsp bulgaricus. Pediococcus acidilactici) by complementation of a LDH-negative mutant of Escherichia coli. The sequence of these genes has revealed the existence of a novel family of NADH-dependent a-hydroxy-acid dehydrogenases which were completely different from the L-LDH family. The structure-function relationships of enzymes from this new family are currently under study via site-directed mutagenesis.


Résumé - Nous avons cloné les gènes de D-lactate-déshydrogénase (D-LDH) et un gène de Dhydroxy- isocaproate-déshydrogénase (D-HicDH) de deux bactéries lactiques (Lactobacillus delbrueckii subsp bulgaricus, Pediococcus acidilacticl) par complémentation d'un mutant LDH-négatif de Escherichia coli. La séquence de ces gènes a révélé l'existence d'une famille de déshydrogénases d'a-hydroxy-acides NADH-dépendantes totalement différente de la famille des L-LDH. Les relations structure-fonction des enzymes de cette nouvelle famille sont actuellement à l'étude par mutagenèse dirigée.
